Background:
Transmembrane activator & calcium modulator and cyclophilin ligand (CAML) interactor (TACI) is lymphocyte specific member1,2 of the tumor necrosis factor-receptor superfamily3 and is expressed in B-lymphocytes and may act to regulate humoral immunity.4 TACI also serves as a receptor for B-cell activating factor of the TNF family (BAFF) and as a proliferation-inducing ligand (APRIL).5

Molecular Weight
The recombinant mature human TACI/Fc is a disulfide-linked homodimeric protein. Based on N-terminal sequencing, the recombinant protein has Ser 2 of human TACI at the amino-terminus. The reduced human TACI/Fc monomer has a calculated molecular mass of 45.2 kDa. In SDS-PAGE, the recombinant TACI/Fc preparation showed an approximately 51 kDa band and an approximately 40 kDa doublet. The presence of multiple bands in the preparation is likely the result of heterogenous glycosylation.

This lyophilized protein is stable for six to twelve months when stored desiccated at -20°C to -70°C. Biological Activity
The biological activity of Human Transmembrane Activator and CAML Interactor is determined by its ability to block human BAFF induced T2B cell survival assay using a concentration range of 1.0-3.0 ug/ml.
Each investigator should determine their own optimal working dilution for specific applications.
