Description
Background:
Interleukin-20 (IL-20) is a pleiotropic inflammatory protein belonging to the IL-10 family of cytokines. Members of this family share partial homology in their amino acid sequences but they are dissimilar in their biological functions. IL-20 is capable of stimulating colony formation by CD34+ multipotential progenitors but not by other progenitor cells (1). It is produced by activated keratinocytes and monocytes and transmits an intracellular signal through two distinct cell-surface receptor complexes on keratinocytes and other epithelial cells. IL-20 regulates proliferation and differentiation of keratinocytes during inflammation, particularly inflammation associated with the skin. A specific receptor for this cytokine is expressed in skin and upregulated dramatically in psoriatic skin, suggesting a role for this protein in epidermal function and psoriasis (2). Human IL-20 is active on mouse cells.
Source
E. coli
Molecular Weight
The predicted molecular weight of Recombinant Human IL-20 is Mr 17.6 kDa.
State of Matter
Lyophilized
Formulation
This recombinant protein was 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) with BSA as a carrier protein. pH 7.2 – 7.3 with no calcium, magnesium, or preservatives.
Purity
>97% by SDS-PAGE and analyzed by silver stain.
Storage and Stability
This lyophilized protein is stable for six to twelve months when stored desiccated at -20°C to -70°C. After aseptic reconstitution, this protein may be stored at 2°C to 8°C for one month or at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. See Product Insert for exact lot specific storage instructions.
Endotoxin
<1.0 EU/µg as determined by the LAL method
Biological Activity
The biological activity of Human IL-20 was determined by its ability to induce proliferation of mouse BaF/3 cells co-transfected with human IL-20 Rα and IL-20 Rβ. The expected ED50 for this effect is typically 0.2 - 0.6 ng/mL.
Each investigator should determine their own optimal working dilution for specific applications.
