Background:
Interleukin-6 (IL-6), also known as BSF2, acts as both a pro-inflammatory and anti-inflammatory cytokine. Produced by T cells, monocytes, fibroblasts, endothelial cells and keratinocytes, IL-6 has diverse biological functions (1). It is secreted by T cells and macrophages to stimulate immune response to trauma, especially burns or other tissue damage leading to inflammation. The role of IL-6 as an anti-inflammatory cytokine is mediated through its inhibitory effects on TNF-alpha and IL-1, and activation of IL-1ra and IL-10. It signals through the IL-6 receptor system that consists of two chains, IL-6R α and gp130. It is up-regulated by IL-1, TNF, PDGF, IFN beta, TNF alpha, NGF, IL-17 and down-regulated by glucocorticoids, IL-4 and TGF beta. IL-6 is also a "myokine," a cytokine produced from muscle and is elevated in response to muscle contraction (2). It stimulates B cell differentiation and antibody production, synergizes with IL-3 in megakaryocyte development and platelet production, induces expression of hepatic acute-phase proteins, and regulates bone metabolism (3-4). Smooth muscle cells in the tunica media of many blood vessels also produce IL-6 as a pro-inflammatory cytokine. It has been shown to suppress the replication of HIV and SIV (5). IL-6 is also essential for hybridoma growth and is found in many supplemental cloning media such as briclone. It is also produced by adipocytes and is thought to be a reason why obese individuals have higher endogeneous levels of CRP. Intranasally administered IL-6 was shown to improve sleep-associated consolidation of emotional memories. Inhibitors of IL-6 are used to treat postmenopausal osteoporosis.

Storage and Stability
This lyophilized protein is stable for six to twelve months when stored desiccated at -20°C to -70°C. After aseptic reconstitution, this protein may be stored at 2°C to 8°C for one month or at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. Biological Activity
The biological activity of Human IL-6 was determined in a cell proliferation assay using a factor dependent murine plasmacytoma cell line, T1165.85.2.1 (Nordan, R.P., Pumphrey, J.G., and B. Rudikoff. 1987. J. Immunol. 139:813). The expected ED50 for this effect is typically 0.2 - 0.8 ng/ml.
Each investigator should determine their own optimal working dilution for specific applications.
