Description
Background:

ST2, also known as IL-1 R4 and T1, is a transmembrane glycoprotein. It belongs to the Interleukin-1 receptor family and is involved in Th2 immune responses (1). ST2 is expressed on mast cells, Th2 cells, macrophages as well as on stressed cardiac myocytes and airway epithelia (2-7). ST2 binds to IL-33, a pro-inflammatory IL-1 family cytokine which induces helper T cells, mast cells, eosinophils, and basophils to produce type 2 cytokines (8-9). IL-33 binding induces the association of ST2 with IL-1 R1 and in macrophages, ST2 interferes with signaling from IL-1 R1 and TLR4 (1, 4). Soluble ST2 can also bind IL-33 to function as a decoy receptor that blocks the ability of IL-33 to signal through transmembrane ST2 (10).

Molecular Weight
The predicted molecular weight of Recombinant Mouse IL-1 R4 is Mr 63 kDa (monomer). However, the actual molecular weight as observed by migration on SDS Page is Mr 73 kDa.

Formulation
This recombinant protein was 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 with no calcium, magnesium, or preservatives.
Purity
>95% by SDS-PAGE and analyzed by silver stain.
Storage and Stability
This lyophilized protein is stable for six to twelve months when stored desiccated at -20°C to -70°C. After aseptic reconstitution, this protein may be stored at 2°C to 8°C for one month or at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. Biological Activity
The biological activity of Mouse IL-1 R4 was determined by its ability to bind rmIL-33 in a functional ELISA type binding assay. The estimated KD is < 50 nM.
Each investigator should determine their own optimal working dilution for specific applications.
