Description
Background:
Interleukin 1α (IL-1α), also called LAF, EP and LEM, is a non-secreted proinflammatory cytokine produced in a variety of cells including monocytes, tissue macrophages, keratinocytes, dendritic cells and T and B cells. The immune regulatory role of IL-1α is exerted on a wide range of cells including lymphocytes, epithelial cells and fibroblasts (1). Both IL-1α and IL-1β share a 25% amino acid sequence homology. They bind to the same receptor and have similar if not identical biological properties (2). These cytokines have a broad range of activities including, stimulation of thymocyte proliferation, by inducing IL-2 release, B cell maturation and proliferation, mitogenic FGF-like activity and the ability to stimulate the release of prostaglandin and collagenase from synovial cells (3). However, whereas IL-1β is a secreted cytokine, IL-1α is predominantly a cell-associated cytokine. IL-1α is also temporarily up regulated during bone formation and the menstrual cycle and can be induced in response to nervous system stimulation (4).

This recombinant protein solution was 0.2 µm filtered and formulated in modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 with no calcium, magnesium, or preservatives present.

>97% by SDS-PAGE and analyzed by silver stain.
Storage and Stability
Working aliquots of human rIL-1 alpha are stable for up to twelve months at -20° C to -70° C in a manual defrost freezer. Upon thawing, rIL-1 alpha, in the presence of a carrier protein, can be stored at 2° - 8°C for one month without detectable loss of activity. For long-term storage, aliquot and freeze at -20° C to -70° C in a manual defrost freezer. Avoid repeated freeze thaw cycles.

Biological Activity
The biological activity of Human IL-1 Alpha was determined in a cell proliferation assay using the murine helper T cell line, D10.G4.1 (Symons, J.A. et al., in Lymphokines and Interferons, a Practical Approach, 1987, M.J. Clemens, A.G. Morris, and A.J.H. Gearing, eds., IRL Press, p. 272). The expected ED50 for this effect is typically 3 - 7 pg/ml.
Each investigator should determine their own optimal working dilution for specific applications.
