Background:
Granulocyte-macrophage colony-stimulating factor (GM-CSF), also known as CSF-2, is a pleiotropic cytokine that controls the production and function of blood cells (1). GM-CSF stimulates stem cells to produce granulocytes (neutrophils, eosinophils and basophils) and monocytes. It is secreted by a variety of cell types including macrophages, T cells, mast cells, endothelial cells and fibroblasts in response to cytokine or inflammatory stimuli (2). The active form of the protein is found extracellularly as a homodimeric receptor complex composed of GM-CSF Rα and the signal transducing common β chain (CD131). GM-CSF inhibits neutrophil migration and enhances the functional activity of the mature end-cells (3). It has a critical role in regulation of surfactant homeostasis and alveolar macrophage innate immune functions in the lung (4). GM-CSF is used in myeloid reconstitution after bone marrow transplantation and following chemotherapy in older adults with acute myelogenous leukemia (5). As a part of the immune/inflammatory cascade, GM-CSF promotes Th1 biased immune response, angiogenesis, allergic inflammation and the development of autoimmunity, therefore, it is worthy of consideration for therapeutic target (6). It has also recently been evaluated in clinical trials for its potential as a vaccine adjuvant in HIV-infected patients.

Formulation
This recombinant protein solution was 0.2 µm filtered and formulated in modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 with no calcium, magnesium, or preservatives present.
Purity
>97% by SDS-PAGE and analyzed by silver stain.
Storage and Stability
This protein is stable for up to twelve months when stored at -20°C to -70°C. After thawing and addition of a carrier protein, this protein may be stored at 2°C to 8°C for one month or for long-term storage, aliquot and store at -20°C to -70°C in a manual defrost freezer. Biological Activity
The biological activity of Human GM-CSF was determined by a cell proliferation assay using the factor-dependent cell line, TF-1 (Kitamura, T. et al., 1989, J. Cell Physiol. 140:323 - 343). The expected ED50 for this effect is typically 6 - 30 pg/ml.
Each investigator should determine their own optimal working dilution for specific applications.
