Specificity:
Rabbit Anti-Human FLICE Inhibitor Protein-α (FLIP-α) recognizes an epitope near the C-terminus of Human, Mouse and Rat FLIP-α. This polyclonal antibody was purified using affinity chromatography.
Background:
Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain (DD)- containing adapter molecules and members of the ICE/CED-3 protease family. Caspases-8 (FLICE) and -10 (FLICE2) are two pivotal members in the ICE/CED-3 protease family. FLICE-inhibitory proteins were identified in virus and human and designated v-FLIPs and FLIP, respectively.1,2 The human FLIP was also cloned by several labs independently and termed Casper, I-FLICE, FLAME-1, CASH and CLARP3-7. FLIP contains two death effector domains (DEDs) and a caspase-like domain. FLIP interacts with adapter protein FADD and caspase-8 and 10, and potently inhibits apoptosis induced by all known death receptors. Four splice variants of c-FLIPs have been identified and termed FLIPα, β, γ, and δ, respectively.

Formulation
This polyclonal antibody is formulated in phosphate buffered saline (PBS) pH 7.4 containing 0.02% sodium azide as a preservative.
Storage and Stability
This polyclonal antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at –20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

Western Blotting: This antibody can be used to detect Human, Mouse and Rat FLIP-α by Western blot analysis at a dilution of 1:500-1:1000 when used in conjunction with compatible secondary reagents, such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190), under either reducing or non-reducing conditions. The positive control for Western blotting is HeLa Whole Cell Lysate (Leinco Prod. No.: H1402).
