Specificity:
Rabbit Anti-Human I-Kappa-B Kinase Epsilon (IKKε) recognizes an epitope near the C-terminus of Human IKKε. This polyclonal antibody was purified using affinity chromatography.
Background:
Nuclear factor kappa B (NF-κB) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NF-κB mediates the expression of a great variety of genes in response to extracellular stimuli. NF-κB is associated with IkB proteins in the cell cytoplasm, which inhibit NF-κB activity. IkB is phosphorylated by IkB kinase (IKK) complex that contains IKKα, IKKβ, and IKKγ. A novel molecule in the IKK complex was recently identified and designated IKKε/IKK-i.1,2 IKKe is required for the activation of NF-κB by PMA and T cell receptors but not by TNFα and IL-1.1 IKKε/IKK-i message is supexpressed in a variety of tissues and is inducible by TNFα, IL-1, and LPS.2

Formulation
This polyclonal antibody is formulated in phosphate buffered saline (PBS) pH 7.4 containing 0.02% sodium azide as a preservative.
Storage and Stability
This polyclonal antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at –20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

Western Blotting: This antibody can be used to detect Human IKKε by Western blot analysis at a concentration of 0.5-1.0 µg/ml when used in conjunction with compatible secondary reagents, such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190), under either reducing or non-reducing conditions. The positive control for Western blotting is Jurkat Whole Cell Lysate (Leinco Prod. No.: J103).
