Description
Specificity:
Rabbit Anti-Human MD-2 recognizes an epitope near the middle region of human, mouse and rat MD-2. This polyclonal antibody was purified using affinity chromatography.

Background:
MD-2 is a member of the Toll/interleukin-1 receptor (TIR)family, a group of proteins that include the Toll-like receptors (TLRs) (1-3). TLRs are signaling molecules that recognize different pathogen-associated molecular patterns (PAMPs) and serve as an important link between the innate and adaptive immune responses (4). TLR4, the major signaling receptor for lipopolysaccharide (LPS), requires the binding of MD-2 to its extracellular region for maximal response to LPS (5,6). The specificity of this response is determined by the species of MD-2; e.g., human MD-2 transfected into mouse cells can cause mouse TLR4 to react to LPS analogs that are normally antagonistic to human but not mouse TLR4 (7).

Formulation
This polyclonal antibody is formulated in phosphate buffered saline (PBS) pH 7.4 containing 0.02% sodium azide as a preservative.
Storage and Stability
This polyclonal antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at –20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

Western Blotting: This antibody can be used to detect Human MD-2 by Western blot analysis at a concentration of 0.5 - 2 µg/ml when used in conjunction with compatible secondary reagents, such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190), under either reducing or non-reducing conditions. The positive controls for Western blotting are Rat Spleen Tissue Lysate (Leinco Prod. No.: R1014) and Mouse Spleen Tissue Lysate (Leinco Prod. No.: M1018).
