Specificity:
Rabbit Anti-Human p53 Upregulated Modulator of Apoptosis (PUMA) recognizes an epitope near the C-terminus of Human and Mouse PUMA. This polyclonal antibody was purified using affinity chromatography.
Background:
Apoptosis is related to many diseases and development. The p53 tumor-suppressor protein induces apoptosis through transcriptional activation of several genes. A novel p53 inducible pro-apoptotic gene was identified recently and designated PUMA (for p53 upregulated modulator of apoptosis) and bbc3 (for Bcl-2 binding component 3) in human and mouse.1-3 PUMA/bbc3 is one of the pro-apoptotic Bcl-2 family members including Bax and Noxa, which are also transcriptional targets of p53. The PUMA gene encodes two BH3 domain-containing proteins termed PUMA-α and PUMA-β.1 PUMA proteins bind Bcl-2, localize to the mitochondria, and induce cytochrome c release and apoptosis in response to p53. PUMA may be a direct mediator of p53-induced apoptosis.

Formulation
This polyclonal antibody is formulated in phosphate buffered saline (PBS) pH 7.4 containing 0.02% sodium azide as a preservative.
Storage and Stability
This polyclonal antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at –20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

IHC (NBF/Par.), WB
Western Blotting: This antibody can be used to detect Human and Mouse PUMA by Western blot analysis at a concentration of 1.0-2.0 µg/ml when used in conjunction with compatible secondary reagents, such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190), under either reducing or non-reducing conditions. The positive controls for Western blotting are K562 Whole Cell Lysate (Leinco Prod. No.: K128) and 3T3 Balb Whole Cell Lysate (Leinco Prod. No.: A183).
