Description
Specificity:
Rabbit Anti-Human TAB1 recognizes human and mouse TAB1. This polyclonal antibody was purified using affinity chromatography.
Background:
TAB1 was identified as a regulator of the MAP kinase kinase kinase TAK1/MAP3K7 (1), which is known to mediate various intracellular signaling pathways, such as those induced by TGF-β and members of the Toll-IL-1R (TIR) superfamily (2), thus acting as an intermediate in both proliferative and innate and adaptive immune responses (3). This protein, together with either TAB2 or TAB3, activates TAK1 kinase in response to upstream signals (4,5). It has been shown that the C-terminal portion of TAB1 is sufficient for binding and activation of TAK1, while a portion of the N-terminus acts as a dominant-negative inhibitor of TGF-β, demonstrating how this protein can function as a mediator between TGF-β receptors and TAK1 (6).

Formulation
This polyclonal antibody is formulated in phosphate buffered saline (PBS) pH 7.4 containing 0.02% sodium azide as a preservative.
Storage and Stability
This polyclonal antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at –20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

Western Blotting: This antibody can be used to detect human and mouse TAB1 by Western blot analysis at a concentration of 0.5 - 1 µg/ml when used in conjunction with compatible secondary reagents, such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190), under either reducing or non-reducing conditions. The positive control for Western blotting is 3T3 Balb Whole Cell Lysate (Leinco Prod. No.: A183).
