Specificity:
Rabbit Anti-Human Beta-Site APP Cleaving Enzyme 2 (BACE2) recognizes an epitope near the N-terminus of human, mouse and rat BACE2. This polyclonal antibody was purified using affinity chromatography.
Background:
Accumulation of the amyloid-β (αβ) plaque in the cerebral cortex is a critical event in the pathogenesis of Alzheimer’s disease. αβ peptide is generated by proteolytic cleavage of the β-amyloid protein precursor (APP) at β- and γ-sites by proteases. The long-sought β-secretase was recently identified by several groups independently and designated β-site APP cleaving enzyme (BACE) and aspartyl protease 2 (Asp2).1-4 A BACE homolog was recently cloned and designated BACE2, Asp1, DRAP (for Down region aspartic protease), and memapsin 1.4-9 BACE2 also cleaves APP at β-site and at a different site within αβ.8 BACE2 locates on chromosome 21q22.3, the so-called ‘Down critical region’, suggesting that BACE2 and αβ may also contribute to the pathogenesis of Down syndrome.6,7

Formulation
This polyclonal antibody is formulated in phosphate buffered saline (PBS) pH 7.4 containing 0.02% sodium azide as a preservative.
Storage and Stability
This polyclonal antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at –20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

Western Blotting: This antibody can be used to detect human, mouse and rat BACE2 by Western blot analysis at a concentration of 0.5 - 1.0 µg/ml when used in conjunction with compatible secondary reagents, such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190), under either reducing or non-reducing conditions. The positive control for Western blotting is Human Brain Tissue Lysate (Leinco Prod. No.: H1407).
