Specificity:
Rabbit Anti-Human Chemokine (C-C Motif) Ligand 23 (CCL23) recognizes Human CCL23. This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research.
Background:
Chemokine (C-C motif) ligand 23 (CCL23) is a small cytokine belonging to the CC chemokine family.1 It is primarily expressed in cells of the myeloid lineage.2 CCL23 induces endothelial cell migration and tube formation and enhances the expression of MMP-2 mRNA and protein levels in endothelial cells. CCL23 may play a direct role in angiogenesis.3

This antigen affinity purified polyclonal antibody is aseptically filtered and lyophilized from PBS with no carrier proteins or preservatives added.
Reconstitution
Aseptically reconstitute this antigen affinity purified polyclonal antibody using sterile deionized water to a concentration >0.2 mg/ml.

Storage and Stability
The lyophilized antibody should be stored desiccated at -20°C. The reconstituted antibody can be stored for at least six weeks at 4°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

Indirect ELISA, N, WB
Indirect ELISA: This antibody can be used in an indirect detection ELISA at 0.5 - 1.0 µg/ml with a suitable second step reagent such as Goat Anti-Rabbit IgG (H&L) Horseradish Peroxidase (Leinco Prod. No.: R115). The detection sensitivity of this indirect ELISA for Human CCL23 is approximately 0.16 ng/well.
Neutralization: This antibody is useful for neutralization of Human CCL23 bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Human CCL23 (at 20 ng/ml) is 0.5 - 2.5 µg/ml.
Western Blotting: To detect Human CCL23 this polyclonal antibody can be used at a concentration of 0.1 - 0.2 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190) and a chromogenic substrate such as TMB HRPO Membrane/IHC Substrate UltraSensitive™ One Component "Ready Use" (Leinco Prod. No.: T343). The detection limit for Human CCL23 is 5 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is Recombinant Human CCL23 (Leinco Prod. No.: M180).
