Specificity:
Rabbit Anti-Human Interleukin-32 (IL-32) recognizes an epitope in the intermediate domain of Human and Mouse IL-32. This polyclonal antibody was purified using affinity chromatography.
Background:
Interleukin-32 (IL-32) was initially identified as a transcript (NK4) that is selectively expressed in lymphocytes and NK cells and whose expression is increased following activation by IL-2 . It was later re-isolated from an IL-18-treated lung carcinoma cell line and re-named IL-32.2 IL-32 is unusual in that it does not share sequence homology with known cytokine families and is highly expressed in immune tissues, existing in at least four differentially spliced isoforms. Because treatment of human monocytic and mouse macrophage cells with IL-32 induces several proinflammatory cytokines such as TNF-α, IL-8 and MIP-2, and because it is also induced in human peripheral lymphocyte cells after mitogen stimulation and in epithelial cells by IFNγ, it has been suggested that IL-32 may play a role in autoimmune and inflammatory diseases such as rheumatoid arthritis.3

Formulation
This polyclonal antibody is formulated in phosphate buffered saline (PBS) pH 7.4 containing 0.02% sodium azide as a preservative.
Storage and Stability
This polyclonal antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at –20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

Western Blotting: To detect Human IL-32 this polyclonal antibody can be used at a concentration of 2.5-5 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190) and a chromogenic substrate such as TMB HRPO Membrane/IHC Substrate UltraSensitive™ One Component "Ready Use" (Leinco Prod. No.: T343). The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.
