Rabbit Anti-Human gAcrp30/Adipolean Variant recognizes Human gAcrp30/Adipolean Variant. This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research.
Background:
Human gAcrp30 is a naturally occurring globular protein, obtained by proteolytic processing of adiponectin. Adiponectin is produced and secreted exclusively by adipocytes, and is a relatively abundant plasma protein, accounting for up to 0.05% of total serum protein. Like Adiponectin, gAcrp30 is capable of decreasing hyperglycemia and reversing insulin resistance. Additionally, gAcrp30 has been shown to be an important factor in promoting fat loss by signaling muscle to absorb and burn Free-Fatty Acids (FFAs). The signaling receptors for adiponectin and gAcrp30 have recently been identified and names AdipoR1 and AdipoR2. AdipoR2 is predominantly expressed in the liver.

Formulation
This antigen affinity purified polyclonal antibody is aseptically filtered and lyophilized from PBS with no carrier proteins or preservatives added.
Reconstitution
Aseptically reconstitute this antigen affinity purified polyclonal antibody using sterile deionized water to a concentration 0.1-1.0 mg/ml.
Storage and Stability
The lyophilized antibody should be stored desiccated at -20°C. The reconstituted antibody can be stored for at least six weeks at 4°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

ELISA Sandwich: This antibody is useful as the capture antibody in a sandwich ELISA. The suggested coating concentration is 0.5-2.0 µg/ml. A suitable detection antibody is Anti-Human gAcrp30/Adipolean Variant-Biotin (Antigen Aff. Pur.) (Leinco Prod. No.: A226) at a concentration of approximately 0.25-1.0 µg/ml. When used in conjunction with an optimal second step reagent such as UltraAvidin™- Horseradish Peroxidase (HRPO) (Leinco Prod. No.: A106) and a chromogenic substrate such as TMB HRPO Microwell Substrate Standard Kinetic One Component "Ready Use" (Leinco Prod. No.: T118), the sensitivity of this sandwich ELISA is 0.2-0.4 ng/well. A suggested standard for this assay is Recombinant Human gAcrp30/Adipolean Variant (Leinco Prod. No.: A227).
Indirect ELISA: This antibody can be used in an indirect detection ELISA at 0.5-2.0 µg/ml with a suitable second step reagent such as Goat Anti-Rabbit IgG (H&L) Horseradish Peroxidase (Leinco Prod. No.: R115). The detection sensitivity of this indirect ELISA for Human gAcrp30/Adipolean Variant is approximately 0.2-0.4 ng/well.
Western Blotting: To detect Human gAcrp30/Adipolean Variant this polyclonal antibody can be used at a concentration of 0.1-0.2 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190) and a chromogenic substrate such as TMB HRPO Membrane/IHC Substrate UltraSensitive™ One Component "Ready Use" (Leinco Prod. No.: T343). The detection limit for Human gAcrp30/Adipolean Variant is 1.5-3.0 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.
