Specificity:
Rabbit Anti-Human Caspase-9 recognizes an epitope in the intermediate domain of Human Caspase-9. This polyclonal antibody was purified using affinity chromatography.
Background:
Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. A novel member in the caspase family was recently identified and designated ICE-LAP6, Mch6, and Apaf-3.1-3 Caspase-9 and Apaf-1 bind to each other, which leads to caspase-9 activation3 Caspase-9 is also activated by granzyme B and CPP32.1,2 Activated caspase-9 cleaves and activates caspase-3 that is one of the key proteases, being responsible for the proteolytic cleavage of many key proteins in apoptosis.3 Caspase-9 play a central role in cell death induced by a wide variety of apoptosis activators including TNFα, TRAIL, anti-CD-95, FADD, and TRADD.4 Caspase-9 is expressed in a variety of human tissues.1,2

Formulation
This polyclonal antibody is formulated in phosphate buffered saline (PBS) pH 7.4 containing 0.02% sodium azide as a preservative.
Storage and Stability
This polyclonal antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at –20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

Western Blotting: This antibody can be used to detect Human Caspase-9 by Western blot analysis at a dilution of 1:1,000-1:2,000 when used in conjunction with compatible secondary reagents, such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190), under either reducing or non-reducing conditions. The positive control for Western blotting is HeLa Whole Cell Lysate (Leinco Prod. No.: H1402).
