Specificity:
Rabbit Anti-Human Casp-4 recognizes an epitope near the N-terminus of mouse and human Casp-4. This polyclonal antibody was purified using affinity chromatography.
Background:
Caspases are a family of cysteine proteases that can be divided into the apoptotic and inflammatory caspase subfamilies. Unlike the apoptotic caspases, members of the inflammatory subfamily are generally not involved in cell death but are associated with the immune response to microbial pathogens.1 Members of this subfamily include caspase-1, -4, -5, and -12. Activation of these caspases results in the cleavage and activation of proinflammatory cytokines such as IL-1β and IL-18.2,3 Caspase-4 was initially identified as a homologous protein to Caspase-1 and the C. elegans Ced-3 which could induce apoptosis in transfected cells.4 More recent studies have shown that it can be activated by ER stress and has been suggested to be involved in multiple neuronal pathologies such as Alzheimer’s disease.5

Formulation
This polyclonal antibody is formulated in phosphate buffered saline (PBS) pH 7.4 containing 0.02% sodium azide as a preservative.
Storage and Stability
This polyclonal antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at –20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

Western Blotting: This antibody can be used to detect mouse and human Casp-4 by Western blot analysis at a concentration of 1 µg/ml when used in conjunction with compatible secondary reagents, such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190), under either reducing or non-reducing conditions. The positive controls for Western blotting are Ramos Whole Cell Lysate (Leinco Prod. No.: R1173) and PN:M1151.
