Specificity:
Rabbit Anti-Human Casp-1 recognizes and epitope in the intermediate domain of Human Casp-1. This polyclonal antibody was purified using affinity chromatography.
Background:
Caspases are a family of cysteine proteases that can be divided into the apoptotic and inflammatory caspase subfamilies. Unlike the apoptotic caspases, members of the inflammatory subfamily are generally not involved in cell death but are associated with the immune response to microbial pathogens.1,2 Members of this subfamily include caspase-1, -4, -5, and -12 and can activate proinflammatory cytokines such as IL-1β and IL-18.3,4 Caspase-1 was initially identified as an IL-1β-converting enzyme;5 later experiments revealed it to be a mammalian homolog of the C. elegans cell death gene ced-3 6 whose overexpression can induce apoptosis in fibroblasts.7

Formulation
This polyclonal antibody is formulated in phosphate buffered saline (PBS) pH 7.4 containing 0.02% sodium azide as a preservative.
Storage and Stability
This polyclonal antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at –20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

Western Blotting: This antibody can be used to detect Human Casp-1 by Western blot analysis at a concentration of 0.5 µg/ml when used in conjunction with compatible secondary reagents, such as Goat Anti-Rabbit IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: R1190), under either reducing or non-reducing conditions. The positive control for Western blotting is Human Heart Tissue Lysate (Leinco Prod. No.: H1405).
