Description
Specificity:
Anti-Human Vascular Endothelial Growth Factor (VEGF) recognizes Human Vascular Endothelial Cell Growth Factor. This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.

Background:
Vascular endothelial growth factor (VEGF), a potent proangiogenic cytokine1 is the key signal used by oxygen-hungry cells to promote growth of blood vessels. It binds to specialized receptors on the surfaces of endothelial cells and directs them to build new vessels.2 VEGF are crucial regulators of vascular development during embryogenesis (vasculogenesis) and blood-vessel formation in the adult (angiogenesis). Abnormal VEGF function is associated with inflammatory diseases including atherosclerosis, and hyperthyroidism.3,4,5,6

Formulation
This purified monoclonal antibody is aseptically filtered and lyophilized from PBS with no carrier proteins or preservatives added.
Reconstitution
Aseptically reconstitute this monoclonal antibody in sterile deionized water to a concentration of 1.0 mg/ml.
Storage and Stability
This lyophilized monoclonal antibody should be stored desiccated at -20°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

ELISA Cap, WB
ELISA Sandwich: This antibody is useful as the capture antibody in a sandwich ELISA. The suggested coating concentration is 2.0 µg/ml. A suitable detection antibody is Anti-Human VEGF-Biotin (Antigen Aff. Pur.) (Leinco Prod. No.: V105). When used in conjunction with an optimal second step reagent such as UltraAvidin™- Horseradish Peroxidase (HRPO) (Leinco Prod. No.: A106) and a chromogenic substrate such as TMB HRPO Microwell Substrate Standard Kinetic One Component "Ready Use" (Leinco Prod. No.: T118), the sensitivity of this sandwich ELISA is 0.1 ng/well. A suggested standard for this assay is Recombinant Human VEGF 165 (Leinco Prod. No.: V101).
Western Blotting: To detect Human VEGF this monoclonal antibody can be used at a concentration of 0.5 µg/ml. This monoclonal antibody should be used in conjunction with compatible second-step reagents such as Goat Anti-Mouse IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: M114) and a chromogenic substrate such as TMB HRPO Membrane/IHC Substrate UltraSensitive™ One Component "Ready Use" (Leinco Prod. No.: T343). The detection limit for Human VEGF is 0.3 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.
