Specificity:
Mouse Anti-Human Endocrine Gland-Derived Endothelial Growth Factor (EG-VEGF) (Clone 188608) recognizes an epitope on Human EG-VEGF. This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.
Antigen Distribution:
Human EG-VEGF is a secreted angiogenetic mitogen growth factor expressed in the steroidogenic glands, ovary, testis, adrenal and placenta.
Background:
Endocrine Gland-Derived Vascular Endothelial Growth Factor (EG-VEGF) or Prokineticin-1 (PK-1) is a novel secreted protein expressed in various tissues including steroidogenic glands. Although structurally non-related to VEGF, EG-VEGF/PK-1 was shown to promote angiogenesis acting via two G-protein coupled receptors (PK-R1 and PK-R2). 1 EG-VEGF is mitogen specific for the endothelium of steroidogenic glands. It induces proliferation, migration and fenestration in capillary endothelial cells derived from endocrine glands. EG-VEGF mRNA expression is localized to the steroid hormone-producing cells, i.e. Leydig cells of the testis and the specialized stroma in the ovary. 2
EG-VEGF increased the phosphorylation of Akt in a phosphatidylinositol 3-kinase-dependent manner and is a survival factor for ACE cells.
EG-VEGF, possibly through binding to a G-protein coupled receptor, results in the activation of MAPK p44/42 and phosphatidylinositol 3-kinase signaling pathways, leading to proliferation, migration, and survival of responsive endothelial cells. 3

Formulation
This monoclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco's phosphate buffered saline (1X PBS) pH 7.2 - 7.3 containing 5.0% w/v trehalose with no calcium, magnesium or preservatives present.
Reconstitution
This lyophilized antibody may be aseptically reconstituted with sterile PBS to a final volume of 1 ml. This will result in a final antibody concentration of 500 µg/ml.
Endotoxin
<0.1 EU/µg as determined by the LAL method
Storage and Stability
The lyophilized antibody can be stored desiccated at -20°C to -70°C for up to twelve months. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid repeated freeze thaw cycles. No detectable loss of activity was observed after six months.

ELISA Cap
ELISA Sandwich: This antibody is useful as the capture antibody in a sandwich ELISA. The suggested coating concentration is 2-8 µg/ml. A suitable detection antibody is Anti-Human EG-VEGF-Biotin (Antigen Aff. Pur.) (Leinco Prod. No.: E147) at a concentration of approximately 0.1-0.4 µg/ml. When used in conjunction with an optimal second step reagent such as UltraAvidin™- Horseradish Peroxidase (HRPO) (Leinco Prod. No.: A106) and a chromogenic substrate such as TMB HRPO Microwell Substrate Standard Kinetic One Component "Ready Use" (Leinco Prod. No.: T118), the sensitivity of this sandwich ELISA is 16 pg/ml. A suggested standard for this assay is Recombinant Human EG-VEGF (Leinco Prod. No.: V108).
