Description

The 5-AMP-activated protein kinase (AMPK) is a member of the SNF1 (sucrose nonfermentor) kinase family (1). AMPK is a heterotrimeric protein comprising alpha (63 kDa), beta (38 kDa) and gamma (38 kDa) subunits (2). The alpha subunit is the catalytic subunit, while beta and gamma are noncatalytic subunits, although they have been found to interact with the active subunit in liver. AMPK regulates fatty acid and sterol synthesis by phosphorylation of acetyl-CoA as well as cholesterol synthesis via phosphorylation and inactivation of hydroxymethylglutaryl-CoA reductase (3). AMPK beta-1 mediates the association of the AMPK heterotrimeric complex in vitro (2). Two isoforms have been found, and the difference in expression patterns indicates tissue-specific roles for these isoforms (4). Phosphorylation on serine 24/25 and serine 182 have been linked to AMPK localization.

A. Western blot analysis on human brain tissue lysate using anti-Phospho-AMPK beta 1(pS181) RabMAb (cat# 2271-1), 1:20,000 dilution. Cells were either (A) untreated (B) treated with Alkaline Phosphatase.

B. Immunohistochemical analysis of paraffin-embedded human skeletal muscle using anti-AMPK RabMAb(cat.#2271-1).

C. Immunofluorescent staining of HeLa cells using anti-AMPK RabMAb (cat. #2271-1).

Specificity

A synthetic phospho-peptide corresponding to residues surrounding Serine 181 of human AMPK beta was used as immunogen.

Storage Buffer & Conditions

50 mM Tris-Glycine (pH 7.4), 0.15 M NaCl, 40% Glycerol, 0.01% sodium azide and 0.05% BSA.
